- Chemiluminescence Immunoassay (CLIA)

Chemiluminescence Immunoassay (CLIA) is a highly sensitive and specific technique that combines chemiluminescence with immunochemical reactions to detect and analyze a wide range of substances such as antigens, antibodies, and enzymes. Due to its high sensitivity, safety, and specificity, CLIA has gained significant popularity in fields like clinical diagnosis, environmental monitoring, and pharmaceutical analysis.

                                                                                                               

Chemiluminescence immunoassay (CLIA) encompasses several methods based on different luminescence mechanisms, including direct chemiluminescence immunoassay, chemiluminescence enzyme immunoassay (CLEIA), and electrochemiluminescence immunoassay. In the chemiluminescence enzyme immunoassay, an enzyme—commonly horseradish peroxidase (HRP)—is conjugated to an antigen or antibody. Following the immunoreaction, the substrate luminol emits light through an oxidation-reduction reaction catalyzed by the enzyme-labeled immune complex and luminescent reagents. The amount of antigen or antibody is then quantified by measuring the intensity of the emitted light.

Although this reaction produces rapid light emission, the luminescence duration is relatively short, which can compromise the stability and reproducibility of detection results. Research has demonstrated that the addition of one or more chemical enhancers can significantly boost the luminescent intensity and extend the duration of light emission.
