 Gentaur RScript™ cDNA Synthesis Kit (Premix + gDNA Remover)

This one-tube master mix combines genomic DNA elimination and reverse transcription in a streamlined, single-step reaction. It efficiently removes genomic DNA while synthesizing high-quality first-strand cDNA, minimizing the risk of sample contamination and RNA degradation commonly caused by complex reagent additions.

Powered by the highly efficient Gentaur RScript II reverse transcriptase, this mix synthesizes cDNA from very low amounts of total RNA or poly(A) mRNA, making it ideal for RNA templates with high GC content and complex secondary structures. The enzyme’s enhanced thermal stability allows reactions up to 65°C, yielding higher cDNA quantities.

The master mix includes a heat-sensitive double-stranded DNase (dsDNase), which removes residual genomic DNA at room temperature without affecting the synthesized cDNA upon inactivation at 50°C. The optimized 5× RScript II All-in-One RT Buffer contains premixed Random Primer and Oligo(dT)18, compatible with both genomic DNA digestion and cDNA synthesis.

Resulting cDNA is fully compatible with dye-based and probe-based qPCR, supporting accurate downstream gene expression analysis.

Figure 1. Reverse transcription efficiency of Gentaur RScript™ cDNA Synthesis Kit evaluated using 100 ng, 10 ng, and 1 ng of HeLa cell RNA templates. Performance was benchmarked against two competitor kits in reverse transcription reactions followed by quantitative PCR (qPCR). The amplification curves demonstrate superior sensitivity and efficiency of Gentaur RScript™ cDNA Synthesis Kit, particularly at low RNA input levels, outperforming competitor reagents.

Figure 2. Using 100 ng of HeLa cell RNA as the template, Gentaur RScript™ cDNA Synthesis Kit was utilized for reverse transcription, followed by cDNA synthesis and quantitative PCR (qPCR). The resulting amplification curve demonstrates that Gentaur RScript™ cDNA Synthesis Kit efficiently eliminates genomic DNA contamination, ensuring high accuracy and precision in amplification. This effective genomic DNA removal prevents interference from residual DNA, leading to reliable and reproducible gene expression quantification results.
